Telescoped Hybrid Batch–Flow Synthesis of 2-Butylbenzofuran
In this article, we describe our team’s approach to the development of an improved synthesis of the pharmaceutically relevant building block 2-butylbenzofuran (BBF). By utilizing a telescoped hybrid batch–flow process composed of three steps, they were able to produce the final product in high yield and purity. The key cyclization step was performed in a continuous process with an integrated workup, making this approach is more efficient compared to the standard batch conditions. The resulting high quality of the product makes crude BBF suitable to be used in the downstream production process of amiodarone without purification. The team used the Design of Experiments approach, which afforded a sustainable process suitable for industrial applications.
